Chronic Hyperglycemia Model

A chronic hyperglycemia model is an experimental system used to reproduce sustained, abnormally high glucose levels and examine their effects on cells, tissues, or developing organisms. Researchers typically maintain elevated glucose exposure over time, allowing glucose-dependent changes in metabolism, cellular signaling, oxidative stress, and inflammatory responses to emerge. In developmental biology, these models help clarify how maternal or embryonic hyperglycemia can influence cell proliferation, differentiation, tissue patterning, and organ formation. They also support evaluation of molecular pathways and potential interventions relevant to diabetes-associated developmental abnormalities, improving understanding of how prolonged metabolic imbalance shapes early development.

This work describes methods to establish acute and chronic hyperglycemia models in zebrafish. The aim is to investigate the impact of hyperglycemia on physiological processes, such as constitutive and injury-induced neurogenesis. The work also highlights the use of zebrafish to follow radiolabeled molecules (here, [18F]-FDG) using PET/CT.

Hyperglycemia is an abnormally high blood glucose level. It is diagnosed by fasting glucose ≥126 mg/dL, 2-hour oral glucose tolerance test (or OGTT) ≥200 mg/dL, random glucose ≥200 mg/dL with symptoms, or HbA1c ≥6.5%.
