Endothelial Cell Generation

Endothelial cell generation is the process of producing endothelial cells, which form the inner lining of blood and lymphatic vessels and regulate vascular development, permeability, and tissue perfusion. In developmental biology, it typically involves the specification and differentiation of mesodermal progenitors or pluripotent stem cells through coordinated signaling cues, followed by selection and maturation of cells expressing endothelial markers. Generated endothelial cells can model vasculogenesis and angiogenesis, support studies of organ development and vascular disease, and provide platforms for drug screening, tissue engineering, and regenerative medicine. Their controlled production also helps investigate how vascular networks form and adapt.

This protocol allows for the reliable generation and characterization of blood outgrowth endothelial cells (BOECs) from a small volume of adult peripheral blood. BOECs can be used as a surrogate for endothelial cells from patients with vascular disorders and as a substrate for the generation of induced pluripotent stem cells.

This work demonstrates a novel approach to assess the proliferation, differentiation, and vessel-forming potential of vascular endothelial stem cells (VESCs) through mammary fat pad transplantation followed by whole-mount tissue preparation for microscopic observation. A lineage tracing strategy to investigate the behavior of VESCs in vivo is also presented.
