Lawn Roller

A lawn roller is an engineering tool that uses a heavy cylindrical drum to press, level, and compact soil or turf surfaces. As the drum rolls, its distributed load applies downward pressure that smooths uneven ground, improves seed-to-soil contact, and helps settle newly installed sod; water-filled or ballast-loaded designs allow the applied force to be adjusted. Lawn rollers are used in landscape construction, sports-field maintenance, and residential lawn establishment, where controlled compaction supports uniform germination and a stable surface. Their design illustrates practical principles of load distribution, rolling resistance, material selection, and mechanical handling.

A Modified Roller Tube Method for the Long-Term Culture of Rodent Brain Slices

This video demonstrates a modified roller tube method for the long-term culturing of brain slices from a rodent model. First, the brain slices are attached to coverslips. Then, flat-sided roller tubes with a hole on the flat side are taken, and the coverslips are positioned on the hole using adhesive discs. Finally, a nutrient medium and a carbon dioxide and air mixture are added to the tube, and the tubes are incubated with a rolling motion.

This article describes a simple, low-cost method of recording the lawn avoidance behavior of Caenorhabditis elegans, using readily available items such as a smartphone and a light emitting diode (LED) light box. We also provide a Python script to process the video file into a format more amenable for counting.

Presented here is a modified roller tube method for culturing and intermittent high-resolution imaging of rodent brain slices over many weeks with precise repositioning on photoetched coverslips. Neuronal viability and slice morphology are well maintained. Applications of this fully enclosed system using viruses for cell-type specific expression are provided.

Electroporation is a commonly employed method for introducing DNA into bacteria in a process known as transformation. Traditional protocols for the preparation of electrocompetent cells are time consuming and labor intensive. This article describes an alternate, rapid, and efficient method for the preparation of electrocompetent cells presently employed by some laboratories.
