Mcf-10a Cells

MCF-10A cells are an immortalized, non-tumorigenic human mammary epithelial cell line used to model normal breast tissue in laboratory research. They grow under defined culture conditions that typically include nutrients and supplements such as epidermal growth factor, insulin, hydrocortisone, and serum, while extracellular matrix environments can guide their organization into three-dimensional acinar structures. In engineering, MCF-10A cells support the design and testing of biomaterials, tissue-engineered breast models, microfluidic systems, and organoid-like cultures. Their ability to form epithelial structures and respond to changes in matrix composition, biochemical signals, and mechanical conditions helps researchers study tissue architecture, cell behavior, and disease-related transformation.

Breast cancer cells exhibit different dielectric properties compared to non-tumor breast epithelial cells. It has been hypothesized that, based on this difference in dielectric properties, the two populations can be separated for immunotherapy purposes. To support this, we model a microfluidic device to sort MCF-7 and MCF-10A cells.

2026

This protocol describes the method for setting up the mammosphere formation assay to study Apoptosis Inhibitor 5 (Api5)- induced cancer stemness in non-tumorigenic breast epithelial cells. Lysates collected from primary generation mammospheres are analysed for different stemness markers to assess the induction of stem cell-like properties.
