Lugol's Staining

Lugol's staining is a microscopy-based method that uses an iodine solution to preserve and visualize microorganisms and particulate biological material in environmental samples. The iodine and potassium iodide formulation penetrates cells and reacts with starch and other cellular components, increasing contrast and helping reveal organism shape, size, and internal structure; starch-rich material typically appears blue-black or dark brown. Researchers apply the stain to water samples, especially for examining phytoplankton, algae, and other planktonic communities, where it supports cell enumeration, taxonomic assessment, and monitoring of aquatic ecosystems. Its consistent contrast also aids environmental surveys and comparisons across sampling sites and seasons.

In order to examine the cellular, structural and molecular layout of tissues and organs, researchers use a method known as histological staining. In this technique, a tissue of interest is preserved using chemical fixatives and sectioned, or cut into very thin slices. A variety of staining techniques are then applied to provide contrast to the visually uniform sections. In the study of neuroanatomy, histological techniques are frequently applied to visualize and study nervous system tissue.
