Absolute Qpcr

Absolute qPCR is a quantitative polymerase chain reaction method that determines the starting amount of a specific DNA target rather than reporting only relative changes, making it valuable for precise genetic measurements. During amplification, target-specific primers and fluorescent dyes or probes generate a signal that increases with each cycle, while the threshold cycle reflects the initial template concentration. Researchers compare this cycle value with a standard curve prepared from samples containing known target quantities to calculate absolute copy number or concentration. In genetics, absolute qPCR supports gene copy-number analysis, transgene validation, mutation detection, and measurement of nucleic acid abundance in biological samples.

Tandem splicing events occur at sites less than 12 nucleotides apart. Quantifying ratios of such splice variants is feasible using an absolute quantitative PCR approach. This manuscript describes how splice variants of the gene STAT3, in which two splicing events results in Serine-701 inclusion/exclusion and α/β C-termini, can be quantified.
