Everted Gut Sac

The Everted Gut Sac is an ex vivo intestinal tissue model used to study how substances cross the gut wall under controlled laboratory conditions. In this technique, a segment of intestine is turned inside out, filled with oxygenated buffer, and immersed in a solution containing the compound of interest; changes inside the sac indicate transport from the mucosal to serosal side. Researchers can measure nutrient, drug, and ion absorption while examining how transporters, epithelial integrity, or genetic variation influence intestinal function. In genetics, the model supports investigation of gene-dependent differences in absorption and helps connect molecular changes with measurable physiological outcomes.

This video illustrates the technique for extracting DNA from the species of microbes resident in the termite hindgut. The preparation of a wet mount slide, which is useful for visualizing the gut microbial community is also illustrated, and a tour through the species-rich gut environment is given.

Dissection of the Endolymphatic Sac from Mice

This study describes how to dissect the endolymphatic sac from the inner ear of the mouse at different ages. The result of a similar dissection is shown in an Slc26a4-null mouse model of enlargement of the endolymphatic sac. A transgenic mouse with a fluorescent reporter expressed in the endolymphatic sac is presented as a model to readily visualize the endolymphatic sac, and educational tool.
