Integrase Strand Transfer Inhibitors

Integrase strand transfer inhibitors are antiretroviral drugs that block HIV from inserting its viral DNA into the genome of an infected host cell, a critical step in establishing infection. They bind the catalytic site of HIV integrase and chelate essential magnesium ions, preventing the strand-transfer reaction that joins viral and cellular DNA. Used in combination antiretroviral therapy, these agents rapidly suppress viral replication and reduce the risk of disease progression and transmission. Their effectiveness also makes them important in immunology and infection research, where drug resistance, adherence, pharmacokinetics, and interactions with host immune responses guide treatment strategies.

A quick and efficient method to integrate foreign DNA of interest into pre-made acceptor strains, termed landing pad strains, is described. The method allows site-specific integration of a DNA cassette into the engineered landing pad locus of a given strain, through conjugation and expression of the ΦC31 integrase.
