Mutant Library

A mutant library is a collection of genetically varied cells, organisms, or gene sequences created to examine how specific mutations alter biological function. Researchers generate variants through targeted or random mutagenesis, introduce them into a suitable system, and screen or select the library under defined conditions to link genotype with phenotype. In immunology and infection research, mutant libraries can reveal pathogen factors that control replication, virulence, immune evasion, or antimicrobial susceptibility, while also supporting studies of antibody and T-cell recognition. These approaches help identify therapeutic targets, clarify host-pathogen interactions, and guide the development of vaccines and diagnostic tools.

The nonredundant transposon insertion mutant library of P. aeruginosa strain PA14, designated as PA14NR Set, facilitates analysis of gene functionality in numerous processes. Presented here is a protocol to generate high-quality copies of the PA14NR Set mutant library.

Construction of a fosmid library with environmental genomic DNA isolated from the vertical depth continuum of a seasonally hypoxic fjord is described. The resulting clone library is picked into 384-well plates and archived for downstream sequencing and functional screening by the application of an automated colony picking system.
