Nanoparticle Labeling

Nanoparticle labeling is the attachment or incorporation of detectable tags onto nanoscale particles, enabling researchers to identify, track, and characterize them in biological systems. Labels such as fluorescent dyes, isotopes, or affinity molecules are introduced by surface adsorption, encapsulation, or covalent coupling, while preserving the particle’s size, composition, and biological function. In immunology and infection research, labeled nanoparticles help visualize interactions with immune cells, monitor cellular uptake and tissue distribution, and evaluate delivery to pathogens or infected sites. These measurements support the development of vaccines, diagnostic probes, antimicrobial delivery systems, and imaging tools, while revealing how nanoparticle design influences immune recognition and therapeutic performance.

Cell Labeling and Targeting with Superparamagnetic Iron Oxide Nanoparticles

Targeted cell delivery is useful in a variety of biomedical applications. The goal of this protocol is to use superparamagnetic iron oxide nanoparticles (SPION) to label cells and thereby enable magnetic cell targeting approaches for a high degree of control over cell delivery and localization.

We describe a technique for labeling and tracking stem cells with FDA-approved, superparamagnetic iron oxide (SPIO), ferumoxytol (Feraheme). This cellular imaging technique that utilizes magnetic resonance (MR) imaging for visualization, is readily accessible for long-term monitoring and diagnosis of successful or unsuccessful stem cell engraftments in patients.

This manuscript describes a novel technique which allows the detection of intracellular gene expression after endocytosis of fluorescence-labeled nanoparticles directly in live cells. The method does not require manipulation of the cells and is not restricted with respect to the target gene or species.
