Tgf Beta Invasion

TGF-beta invasion refers to the ability of transforming growth factor beta signaling to promote cell movement through surrounding tissues, a process relevant to cancer progression and tissue remodeling. After TGF-beta binds its receptors, receptor serine/threonine kinase activity activates Smad-dependent and other intracellular pathways that can induce epithelial-to-mesenchymal transition, cytoskeletal changes, extracellular matrix remodeling, and increased cell motility. In medicine, studying this process helps explain how tumors invade local tissue and spread to distant sites. Experimental models of TGF-beta invasion also support the evaluation of signaling inhibitors, prognostic biomarkers, and therapeutic strategies aimed at limiting metastasis.

An assay to quantitatively measure Transforming Growth Factor (TGF)-β-induced invasion in 3-dimensional collagen gels is described. This assay takes advantage of the MCF10A series of cell lines, which represent different stages of breast cancer development. This method can be adopted to be used with other cell lines and might be used to investigate other potential activators or inhibitors of invasion.

Herein we report a tailored HPLC purification protocol that yields high-purity amyloid beta 42 (Aβ42) and amyloid beta 40 (Aβ40) peptides, capable of oligomer formation. Amyloid beta is a highly aggregation prone, hydrophobic peptide implicated in Alzheimer's disease. The amyloidogenic nature of the peptide makes its purification a challenge.
