Glycolytic Flux

Glycolytic flux is the rate at which glucose-derived carbon moves through the glycolysis pathway, a central measure of how cells balance energy production and metabolic demand. It depends on glucose availability and the activity of rate-controlling enzymes, which convert glucose to pyruvate while generating ATP and NADH; pyruvate can then enter mitochondria or be reduced to lactate. In neuroscience, measuring glycolytic flux helps distinguish how neurons and glial cells fuel activity, respond to hypoxia or injury, and coordinate metabolic support, providing insight into brain energetics, neurodegeneration, and potential therapeutic targets.

Glycolysis is a defining metabolic marker in multiple biological systems. Monitoring glycolysis by measuring the extracellular flux of H+ is common, but requires correction to be quantitative and unambiguous. Here, we demonstrate how to gather and correct extracellular flux data to distinguish between respiratory and glycolytic sources of extracellular acidification.

Metabolic reprogramming is a characteristic and prerequisite for M1 and M2 macrophage polarization. This manuscript describes an assay for the measurement of fundamental parameters of glycolysis and mitochondrial function in mouse bone marrow-derived macrophages. This tool can be applied to investigate how particular factors affect the macrophage’s metabolism and phenotype.
