Hippocampal Dendrites

Hippocampal dendrites are branched neuronal extensions that receive and integrate synaptic signals within the hippocampus, a brain region essential for learning and memory. Their arborization, dendritic spines, and ion channels shape how excitatory and inhibitory inputs combine through spatial and temporal summation, while activity-dependent changes such as long-term potentiation can strengthen selected synapses. Studying these structures helps explain how neural circuits encode spatial information, form memories, and adapt to experience. Dendritic morphology and synaptic function are also investigated in models of aging, epilepsy, stress, and neurodegenerative disease, providing insight into how altered connectivity affects hippocampal computation and behavior.

Assessment of Dendritic Arborization in the Dentate Gyrus of the Hippocampal Region in Mice

We describe two methods for visualization and quantification of dendritic arborization in the hippocampus of mouse models: real-time and extended depth of field imaging. While the former method allows sophisticated topographical tracing and quantification of the extent of branching, the latter allows speedy visualization of the dendritic tree.

Imaging Dendritic Spines of Rat Primary Hippocampal Neurons using Structured Illumination Microscopy

This article describes a working protocol to image dendritic spines from hippocampal neurons in vitro using Structured Illumination Microscopy (SIM). Super-resolution microscopy using SIM provides image resolution significantly beyond the light diffraction limit in all three spatial dimensions, allowing the imaging of individual dendritic spines with improved detail.

FRAP has been used to quantify the mobility of Green Fluorescence Protein (GFP)-tagged proteins in cultured cells. We examined the mobile/immobile fractions of the GFP by analyzing the fluorescence recovery percentage after photobleaching. In this study, FRAP was performed at spines of hippocampal neurons.

This protocol outlines the steps required to dissect, transfect via electroporation and culture mouse hippocampal and cortical neurons. Short-term cultures may be used for studies of axon outgrowth and guidance, while long-term cultures can be used for studies of synaptogenesis and dendritic spine analysis.
