Tissue Slide Preparation

Tissue slide preparation is a laboratory process that converts biological tissue into thin, preserved sections for microscopic examination, enabling researchers to study cellular structure and organization. In neuroscience, tissue is typically fixed to preserve morphology, embedded or frozen for support, sectioned with a microtome or cryostat, and mounted on glass slides before staining or antibody labeling reveals neurons, glia, fibers, or molecular markers. Careful control of fixation, section thickness, and staining preserves anatomical detail and improves signal quality. Prepared slides support the analysis of brain architecture, neural connectivity, disease-related changes, and responses to experimental treatments.

This manuscript presents a protocol for the histological preparation of slides from rodent eyeballs. With the proposed method, the inner retina can be easily visualized and assessed under a light microscope. The procedure is presented for the eyeballs of mice and rats.
