Glutamine Supplemented Diet

A glutamine-supplemented diet is a feeding regimen that adds glutamine, a conditionally essential amino acid, to standard food or drinking solutions to test how nutrition influences physiology and behavior. After ingestion, glutamine is absorbed and metabolized as a source of carbon and nitrogen; in neural and peripheral tissues, it can also contribute to production of the neurotransmitter precursors glutamate and γ-aminobutyric acid (GABA). Behavioral studies use this approach to assess whether altered amino-acid availability affects activity, learning, stress-related responses, or social behavior. Findings can connect dietary composition with metabolic, neural, and behavioral regulation while informing nutritional models of brain function.

Here we describe a common method to induce chronic liver injury in mice by feeding of a choline-deficient and ethionine-supplemented (CDE) diet. We demonstrate health monitoring, liver perfusion, isolation, and preservation. A time course of six weeks can inform about liver injury, pathohistology, fibrosis, inflammatory, and liver progenitor cell responses.

This article will demonstrate how to monitor glutamine dynamics in live cells using FRET. Genetically encoded sensors allow real-time monitoring of biological molecules at a subcellular resolution. Experimental design, technical details of the experimental settings, and considerations for post-experimental analyses will be discussed for genetically encoded glutamine sensors.

Current treatments of inflammatory bowel diseases (IBD) aim at alleviating disease symptoms but may cause severe side effects. Thus, alternative strategies are being investigated in animal colitis models. Here, we explain how the beneficial effects of diet supplements on clinical IBD signs are analyzed in such a colitis model.
