Sequential Extraction

Sequential extraction is a method that separates compounds from a complex sample by applying multiple extraction steps in sequence, often with solvents of different polarities. Each solvent selectively dissolves and removes compounds according to their solubility, while the remaining material undergoes subsequent treatments. In biochemistry, this approach can fractionate metabolites, lipids, pigments, and other biomolecules before qualitative or quantitative analysis. By reducing sample complexity and enriching chemical classes, sequential extraction supports chromatography, structure elucidation, bioactivity screening, and studies of biochemical composition.

Sequential salt extraction of chromatin bound proteins is a useful tool for determining the binding properties of large protein complexes. This method can be employed to evaluate the role of individual subunits or domains in the overall affinity of a protein complex to bulk chromatin.

Here, we present a protocol for the isolation of increasingly insoluble/aggregated alpha-synuclein (α-syn) from post-mortem human brain tissue. Through the utilization of buffers with increasing detergent strength and high-speed ultracentrifugation techniques, the variable properties of α-syn aggregation in diseased and non-diseased tissue can be examined.

Electricity-Free, Sequential Nucleic Acid and Protein Isolation

A tool and chemistries are described to sequentially isolate nucleic acids followed by proteins from a sample without the need for electricity. The tool consists of a sorbent held within a transfer pipette while the isolation chemistries are based on solid-phase extraction principles. The isolated macromolecules can be analyzed by immuno-based and PCR-based assays.
