Acceptor Sensitized Emission

Acceptor sensitized emission is a fluorescence method used to detect molecular proximity and interactions by measuring light emitted from an acceptor fluorophore after excitation of a donor. When donor and acceptor labels lie within nanometer-scale distance, the donor transfers excitation energy nonradiatively to the acceptor through Förster resonance energy transfer, causing acceptor emission at a longer wavelength. Quantifying this signal typically requires correction for direct acceptor excitation and donor fluorescence leaking into the detection channel. In biology, the method helps analyze protein associations, conformational changes, biosensor responses, and dynamic interactions in cells and other biological samples.

The synthesis of fluorine-18 (18F) labeled radiopharmaceuticals for positron emission tomography typically requires months of experience. When incorporated into a radiotracer, the silicon-fluoride acceptor (SiFA) motif enables a simple 18F-labeling protocol that is independent of costly equipment and preparatory training, while reducing precursor quantity needed and utilizing milder reaction conditions.
