Adult Zebrafish Brain

The adult zebrafish brain is the mature central nervous system of Danio rerio, a vertebrate model used to study neural structure, function, and disease. It contains organized regions and neural circuits that process sensory information and regulate behavior, while retaining a notable capacity for adult neurogenesis and regeneration, in which neural progenitor cells generate new neurons and support repair after injury. Researchers use this model to examine neuronal regeneration, behavior, brain development, and responses to genetic or pharmacological manipulation, providing comparative insight into vertebrate neuroscience and mechanisms relevant to human neurological disorders.

Here we provide a reproducible method to examine adult neurogenesis using a neurosphere assay derived from the whole brain or from either the telencephalic, tectal or cerebellar regions of the adult zebrafish brain. Additionally, we describe the procedure to manipulate gene expression in zebrafish neurospheres.

In this article, we demonstrate a method for manipulation of gene expression in the ventricular cells of the adult zebrafish telencephalon using antisense morpholino oligonucleotides. We present this method as an efficient and quick protocol that can be used for functional studies in the adult vertebrate brain.
