Allele Quantification

Allele quantification is the measurement of the relative or absolute abundance of alternative forms of a gene within a biological sample, helping characterize genetic variation and its distribution. The process distinguishes alleles through sequence-specific differences, then estimates their abundance using approaches such as allele-specific amplification or sequencing read counts, often normalized against total genetic signal. In biology, allele quantification supports analysis of heterozygosity, allele-specific expression, genetic mosaicism, and changes in variant frequency across samples. These measurements help researchers connect genotype with molecular phenotype, track genetic changes, and assess population or disease-associated variation.

Genetic associations often remain unexplained at a functional level. This method aims to assess the effect of phenotype-associated genetic markers on gene expression by analyzing cells heterozygous for transcribed SNPs.
