Odor Dilution

Odor dilution is the controlled reduction of an odorant’s concentration to measure how its intensity or detectability changes, making it useful for studying chemical communication and sensory perception in biology. Researchers typically prepare serial dilutions by mixing an odor source with an odor-free solvent or carrier, then compare responses as molecules become less available to olfactory receptors and detection approaches an individual’s sensory threshold. This approach supports odor threshold testing, behavioral assays, and investigations of how animals locate food, recognize mates, avoid threats, or respond to environmental chemicals. Quantifying dilution responses can link odor concentration with neural and behavioral outcomes.

This manuscript describes a protocol to examine the olfactory system of rodents. The olfactory habituation/dishabituation test will allow investigators to determine whether a mouse habituates to a repeatedly presented odor and whether the mouse demonstrates dishabituation when presented a novel odor.

In this video article, we describe a new method allowing the construction of odorant gradients with stable and controllable geometries. We briefly illustrate how these gradients can be used to screen for olfactory defects (full and partial anosmia) and to study more subtle features of chemotaxis behavior.

Solution Concentration and Dilution

The relative amount of a given solution component is known as its concentration. Often, though not always, a solution contains one component with a concentration that is significantly greater than that of all other components. This component is called the solvent and may be viewed as the medium in which the other components are dispersed or dissolved. Solutions in which water is the solvent are, of course, very common on our planet. A solution in which water is the solvent is called an aqueous...

Real-Time Monitoring of Odorant Receptor Activation in Recombinant Cells

(2019)This video demonstrates the method to monitor the real-time activation of odorant receptors in recombinant mammalian cells using luminescence measurement. Odorant binding triggers G-protein activation and cAMP production, activating glosensor proteins that emit luminescence upon substrate binding, providing a quantitative measure of receptor response.
