Bacmid Viral Genome

A bacmid viral genome is a large, engineered baculovirus DNA molecule maintained as a bacterial artificial chromosome (BAC) in Escherichia coli, combining viral genetic information with bacterial cloning features. In a typical system, a gene of interest is inserted into a designated attTn7 site through transposon-mediated recombination, while antibiotic resistance and other markers support selection and verification in bacteria. The purified recombinant bacmid is then introduced into insect cells, where it produces infectious recombinant baculovirus particles and directs expression of the inserted gene. This platform supports protein production, gene-function studies, vaccine development, and the construction of complex genetic tools in molecular and cell biology.

Begin with a BACmid, a bacterial artificial chromosome that can replicate like a plasmid inside bacteria. The BACmid contains the entire genome of the vaccinia virus with targeted modifications, but it is not infectious as it requires viral proteins to function.Take a helper virus, which is an attenuated vaccinia virus encoding the gene for EGFP.Infect epithelial cells with the helper virus and simultaneously transfect with the BACmid. The virus expresses EGFP and provides the viral proteins,...

Size and Structure of Viral Genomes

2025

Viral genomes exhibit remarkable diversity in size, structure, and composition, influencing their replication strategies and interactions with host cells. These genomes consist of either DNA or RNA and may be linear or circular. Additionally, they can be single-stranded or double-stranded, with each configuration affecting how the virus propagates within a host. RNA viruses, for instance, generally have smaller genomes than DNA viruses, a factor that contributes to their high mutation rates and...

Single Virus Genomics (SVG) is a method to isolate and amplify the genomes of single virons. Viral suspensions of a mixed assemblage are sorted using flow cytometry onto a microscope slide with discrete wells containing agarose, thereby capturing the virion and reducing genome shearing during downstream processing. Whole genome amplification is achieved using multiple displacement amplification (MDA) resulting in genomic material that is suitable for sequencing.

Nested-PCR to Detect a Specific Viral Genomic Sequence

This video describes nested polymerase chain reaction, a technique that consists of two sequential PCR amplification processes using two primer sets. The first set of primers is intended to anneal to sequences upstream of the second set, resulting in selective amplification of specific gene sequences. This PCR is more sensitive and specific than a normal PCR and is widely used as a detection technique for various diseases.
