Murine Skin Wound

A murine skin wound is a controlled injury to the skin of a mouse, used to study tissue repair, inflammation, and regeneration in a living organism. Healing proceeds through overlapping phases: hemostasis limits blood loss, inflammatory cells remove damaged material, keratinocytes and fibroblasts rebuild tissue, and remodeling strengthens the extracellular matrix. Researchers create standardized wounds and monitor closure, re-epithelialization, granulation tissue, angiogenesis, and scar formation over time. This model helps reveal how genes, immune signals, therapies, and disease conditions influence wound healing, providing an accessible system for investigating repair mechanisms and evaluating potential treatments relevant to human skin injury.

The rat, due to its size, availability, and rather docile behavior, has been utilized as a research model for many years. The goal of this protocol is to utilize the rat as an ischemic skin wound healing model to provide valuable insight into the pathophysiology of chronic wounds.

A Murine Skin Wound Infection Model to Study the Immune Response against a Bacterial Pathogen

This video demonstrates a technique to generate a murine model of skin wound infection. Upon infecting mice — engineered to express a fluorescent protein in neutrophils — with a genetically modified bioluminescent Staphylococcus aureus — a pathogenic bacteria. The bacterial burden at the infection site and the neutrophil recruitment dynamics can be studied using whole animal imaging.

The aim of this study was to develop a murine model of burn wound healing. A thermal burn was induced on the dorsal skin of mice using a preheated brass template. Burned tissue was debrided and overlaid with a skin graft harvested from the tail of a genetically similar donor mouse.
