Biochemistry

Biochemistry is the study of the chemical molecules and reactions that sustain life, linking molecular structure to biological function. It examines how proteins, carbohydrates, lipids, nucleic acids, and water interact through regulated pathways, often with enzymes lowering activation energy and controlling reaction rates under cellular conditions. By analyzing metabolism, energy transfer through ATP, gene expression, and molecular interactions, biochemistry explains how cells grow, communicate, maintain homeostasis, and respond to their environments. Its principles support research in genetics, physiology, microbiology, pharmacology, and biotechnology, helping researchers investigate disease mechanisms, identify therapeutic targets, and develop diagnostic or industrial applications.

Alterations in the kynurenine pathway (KP) neuroactive metabolites are implicated in psychiatric illnesses. Investigating the functional outcomes of an altered kynurenine pathway metabolism in vivo in rodents may help elucidate novel therapeutic approaches. The current protocol combines biochemical and behavioral approaches to investigate the impact of an acute kynurenine challenge in rats.

Here we present a step-by-step protocol of the long-length electrostatic repulsion-hydrophilic interaction chromatography-tandem mass spectrometry (LERLIC-MS/MS) method. This is a novel methodology that enables for the first time quantification and characterization of the glutamine and asparagine deamidation isoforms by shotgun proteomics.

Injection of An. stephensi Embryos to Generate Malaria-resistant Mosquitoes

Anopheles stephensi mosquitoes are vectors for malaria inhabiting India and throughout Asia. This video demonstrates the technique for performing microinjections of this species with transgenes that will confer resistance to the malaria to the mosquito. Much of the methodology demonstrated in this video is applicable to microinjection techniques of other mosquito species.

The accuracy and sensitivity of protein determination by the rapid and convenient Bradford assay is compromised by intrinsic nonlinearity. We show a simple linearization procedure that greatly increases the accuracy, improves the sensitivity of the assay about 10-fold, and significantly reduces interference by detergents.
