Chemical Cleavage Method

Chemical cleavage method is a laboratory strategy for breaking selected covalent bonds in biological molecules, producing smaller fragments that can be analyzed or used in downstream experiments. The process relies on a chemical reagent reacting with a susceptible bond under controlled conditions, with factors such as molecular structure, pH, solvent, temperature, and reaction time influencing cleavage specificity and efficiency. In biology, chemical cleavage helps fragment proteins, peptides, or nucleic acids for sequence analysis, structural mapping, and identification of functional regions. By generating interpretable products from complex molecules, the method supports biomolecular characterization and complements enzymatic digestion, separation techniques, and other analytical approaches.

This video demonstrates an assay to screen for the proteolytic activity of proteases using fluorogenic peptides. The protease recognizes its cleavage site on the peptide, cleaving it and separating the quencher from the fluorophore, enabling its fluorescence emission. The fluorescence signal is detected and analyzed to check for the cleavage efficiency of different peptide variants.

We have engineered the capsid protein of hepatitis E virus as a theranostic nanoparticle (HEVNP). HEVNP self-assembles into a stable icosahedral cage in mucosal delivery. Here, we describe the modification of HEVNPs for tumor targeting by mutating surface-exposed residues to cysteines, which conjugate synthetic ligands that specifically bind tumor cells.
