Embryo Mounting

Embryo mounting is a microscopy preparation technique that positions and immobilizes embryos on a slide or imaging dish so their structure, development, and cellular organization can be examined. Researchers typically orient the specimen in a supportive medium, often low-melting-point agarose, which stabilizes the embryo while preserving access to transmitted light or fluorescence imaging. Careful positioning and specimen integrity are essential for visualizing tissues, organ formation, cell movements, and developmental changes across time. The method supports reproducible microscopy in developmental biology, embryology, and disease models, enabling researchers to compare phenotypes and document dynamic processes in living or fixed embryos.

The zebrafish embryo is an excellent model for developmental biology research. During embryogenesis, zebrafish develop with a yolk mass, which presents three-dimensional challenges for sample observation and analysis. This protocol describes how to create two-dimensional flat mount preparations of whole mount in situ (WISH) stained zebrafish embryo specimens.
