Endothelial Cell Proliferation

Endothelial cell proliferation is the process by which endothelial cells divide to expand the cellular lining of blood vessels, supporting vascular growth and repair. In response to signals such as vascular endothelial growth factor, cells progress through the cell cycle, replicate their DNA, and undergo mitosis, often alongside migration and organization into new vessel structures. Studying this process helps explain angiogenesis during development, wound healing, and disease. In laboratory biology, proliferation assays and cultured endothelial cells are used to evaluate vascular responses, investigate cardiovascular disorders, and assess how drugs or biomaterials influence blood vessel formation.

Restenosis following cardiovascular procedures (bypass surgery, angioplasty, or stenting) is a significant problem reducing the durability of these procedures. An ideal therapy would inhibit smooth muscle cell (VSMC) proliferation while promoting regeneration of the endothelium. We describe a model for simultaneous assessment of VSMC proliferation and endothelial function in vivo.

We provide a method for isolating and culturing pure populations of heart valve endothelial cells (VEC). VEC can be isolated from either side of the cusp or leaflet and immediately following, underlying interstitial cell (VIC) isolation is straightforward.
