Epigenetic Histone Modifications

Epigenetic histone modifications are reversible chemical changes to histone proteins that regulate how DNA is packaged and interpreted without altering its nucleotide sequence. Enzymes called writers add marks such as acetyl, methyl, phosphate, or ubiquitin groups to histone tails, while erasers remove them and reader proteins recognize their patterns. These changes can alter chromatin accessibility, influencing whether genes are available for transcription; for example, histone acetylation is often associated with more open chromatin, whereas methylation can activate or repress genes depending on its location. Studying these modifications helps explain cell differentiation, development, disease mechanisms, and potential targets for epigenetic therapies.

The histone proteins have a flexible N-terminal tail extending out from the nucleosome. These histone tails are often subjected to post-translational modifications such as acetylation, methylation, phosphorylation, and ubiquitination. Particular combinations of these modifications form “histone codes” that influence the chromatin folding and tissue-specific gene expression. Acetylation The enzyme histone acetyltransferase adds acetyl group to the histones. Another enzyme, histone deacetylase,...

A reliable and useful approach to detect histone modifications on specific plant genes is described. The approach combines chromatin immunoprecipitation (ChIP) and real-time quantitative PCR. It allows detection of histone modifications on specific genes with a role in diverse physiological processes.

An Overview of Epigenetics

2023

Since the early days of genetics research, scientists have noted certain heritable phenotypic differences that are not due to differences in the nucleotide sequence of DNA. Current evidence suggests that these “epigenetic” phenomena might be controlled by a number of mechanisms, including the modification of DNA cytosine bases with methyl groups, the addition of various chemical groups to histone proteins, and the recruitment of protein factors to specific DNA sites via interactions with...
