Indirect Immunofluorescence Assay

Indirect immunofluorescence assay (IFA) is an antibody-based method for detecting and localizing specific antigens or antibodies in fixed cells, tissues, or clinical samples. In the assay, a primary antibody binds the target, and a fluorophore-labeled secondary antibody recognizes the primary antibody, producing a visible signal under a fluorescence microscope. Because multiple secondary antibodies can bind one primary antibody, indirect immunofluorescence assay can increase signal intensity while allowing one labeled secondary reagent to detect different primary antibodies from the same host species. Researchers and clinicians use IFA to examine protein distribution, characterize cell structures, detect pathogen-specific antibodies, and support autoimmune disease investigation.

Comet Assay as an Indirect Measure of Systemic Oxidative Stress

Comet assay measures DNA breaks, induced by different factors. If all factors (except oxidative stress) causing DNA damage are kept constant, the amount of DNA damage is a good indirect parameter of oxidative stress. The goal of this protocol is to use comet assay for indirect measurement of oxidative stress.

The indirect immunofluorescence protocol described in this article allows the detection and the localization of proteins in the mouse mammary gland. A complete method is given to prepare mammary gland samples, to perform immunohistochemistry, to image the tissue sections by fluorescence microscopy, and to reconstruct images.
