Forelimb Hindlimb Function

Forelimb hindlimb function describes how the paired limbs of vertebrates perform movement, support, and interaction with the environment, reflecting adaptations to different modes of locomotion. Forelimbs commonly contribute to steering, grasping, climbing, or weight bearing, while hindlimbs often generate propulsion through coordinated muscle contraction, joint movement, and skeletal leverage; however, their roles vary among species. Surgical induction of hindlimb ischemia in the mouse is useful to examine angiogenesis, however this is compromised in certain inbred mouse strains that display marked ischemia-induced tissue necrosis. Methods are described to induce subacute limb ischemia using ameroid constrictors to circumvent this problem through the induction of gradual arterial occlusion.

We present a reproducible mouse model of heterotopic hindlimb-to-neck vascularized composite allotransplantation. This supermicrosurgical protocol, involving 0.3-0.5 mm vessel anastomoses, integrates nylon filament guidance and venous cuffing. It provides a reliable platform for translational studies on tolerance, ischemia-reperfusion injury, and immunological mechanisms in vascularized composite allotransplantation.
