Β-galactosidase Assay

A β-galactosidase assay is a laboratory method for measuring β-galactosidase activity, an enzyme that hydrolyzes β-galactosides and serves as an important reporter of gene expression in biology. In a common colorimetric format, the enzyme cleaves the substrate o-nitrophenyl-β-D-galactopyranoside (ONPG) to produce yellow o-nitrophenol, allowing activity to be quantified by measuring absorbance. Other substrates, such as X-gal, generate a blue product for visual detection. These assays help researchers study the lac operon, promoter activity, transcriptional regulation, and cellular responses, while providing a sensitive, accessible approach for comparing gene expression across experimental conditions.

In this video, we demonstrate the modified yeast one-hybrid assay, which detects multiple proteins interacting with target DNA. The transformed yeast cells contain the target DNA sequence upstream of the reporter gene. Additionally, they express transcription factor proteins. When the proteins form complexes and bind to the target DNA, the reporter gene is expressed and can be detected enzymatically.
