Hematopoietic Differentiation

Hematopoietic differentiation is the biological process by which hematopoietic stem cells and progenitor cells develop into the diverse blood cell types required for oxygen transport, immune defense, and hemostasis. It proceeds through progressive lineage commitment, as changes in gene expression, transcription factor activity, and signals from the bone marrow microenvironment guide cells toward erythroid, myeloid, or lymphoid fates. Studying this process clarifies how blood formation is maintained throughout life and how abnormalities arise in anemia, leukemia, and immune disorders. In research, hematopoietic differentiation models support the study of stem cell biology, disease mechanisms, blood development, and potential cell-based therapies.

Here, we present human pluripotent stem cell (hPSC) culture protocols, used to differentiate hPSCs into CD34+ hematopoietic progenitors. This method uses stage-specific manipulation of canonical WNT signaling to specify cells exclusively to either the definitive or primitive hematopoietic program.

An ex vivo protocol to generate mature human red blood cells from hematopoietic stem/progenitors is described. Additionally we describe an efficient lentiviral-delivery method to knockdown the transcription factor TAL1 in primary erythroid cells. The efficiency of lentivirus mediated gene delivery is demonstrated using GFP expressing viruses.

The colony forming cell (CFC) assay is an in vitro assay in which hematopoietic progenitors form colonies in a semi-solid medium. A combination of colony morphology, cell morphology, and flow cytometry are used to assess the ability of the progenitors to proliferate and differentiate along the different hematopoietic lineages.
