Human Dental Epithelial Cells

Human dental epithelial cells are specialized epithelial cells associated with tooth-forming tissues and the oral cavity, where they support tooth development, tissue organization, and epithelial barrier functions. Their behavior depends on cell polarity, cell-cell adhesion, interactions with the basement membrane, and signaling pathways that regulate proliferation and differentiation. In laboratory culture, these cells provide models for investigating odontogenesis, enamel-forming cell development, and epithelial responses to injury or disease. They also support research in dental tissue engineering and regenerative biology, where controlled epithelial-mesenchymal interactions may help guide the formation of tooth-like structures and improve understanding of oral health.

The method described isolation and characterization of human Dental Pulp Stem Cells (hDPSCs) by using either enzymatic dissociation of pulp (DPSC-ED) or direct outgrowth of stem cells from pulp tissue explants (DPSC-OG). Then followed by in vitro comparative differentiation of both types of hDPSCs into odontoblasts.

Here we describe a detailed method for growing primary human bronchial epithelial cells from explants of human bronchial airway tissue including differentiated growth on an air-liquid interface. This method provides an abundant source of primary cells for investigating the role of the airway epithelium in human lung health and disease.
