Human Umbilical Vein Ec

Human umbilical vein endothelial cells (HUVECs) are vascular cells isolated from the inner lining of the umbilical vein and widely used as an in vitro model of human blood vessels. When cultured under controlled conditions, they adhere to extracellular matrix, form a cobblestone-like monolayer, and respond to vascular signals by regulating permeability, migration, and new vessel formation. In biology, HUVECs support studies of endothelial barrier function, angiogenesis, inflammation, and cell signaling. They also provide a practical platform for evaluating biomaterials, vascular therapies, and drug effects, although their neonatal origin and culture conditions should be considered when interpreting results.

This video protocol illustrates the isolation and culture of human umbilical vein endothelial cells (HUVEC) from human umbilical cord. Once isolated these cells can be used for in vitro angiogenesis assays like the Optimized Fibrin Gel Bead Assay also demonstrated by the Hughes lab.

This article first describes a procedure for isolating human endothelial cells from umbilical veins and then shows how to use these cells to examine neutrophil transmigration under flow conditions. By using a low-volume flow chamber made from a polymer with the optical characteristics of glass, live-cell fluorescent imaging of rare cell populations is also possible.

The goal of this protocol is to isolate endothelial progenitor cells from umbilical cord blood. Some of the applications include using these cells as a biomarker for identifying patients with cardiovascular risk, treating ischemic diseases, and creating tissue-engineered vascular and heart valve constructs.
