Hyperglycemia

Hyperglycemia is a condition in which blood glucose levels rise above the normal physiological range, making it an important subject in biology and metabolic research. It occurs when insulin production or action is insufficient to promote glucose uptake by cells, or when glucose production by the liver exceeds cellular use, disrupting energy balance and osmotic regulation. Studying hyperglycemia helps researchers understand glucose homeostasis, pancreatic beta-cell function, insulin resistance, and diabetes-related changes in tissues. Biological models and clinical measurements use these principles to investigate disease mechanisms, evaluate treatments, and clarify how prolonged high glucose can impair cellular and organ function.

This work describes methods to establish acute and chronic hyperglycemia models in zebrafish. The aim is to investigate the impact of hyperglycemia on physiological processes, such as constitutive and injury-induced neurogenesis. The work also highlights the use of zebrafish to follow radiolabeled molecules (here, [18F]-FDG) using PET/CT.

Hyperglycemia is an abnormally high blood glucose level. It is diagnosed by fasting glucose ≥126 mg/dL, 2-hour oral glucose tolerance test (or OGTT) ≥200 mg/dL, random glucose ≥200 mg/dL with symptoms, or HbA1c ≥6.5%. When compared to the previous paper protocol, implementation of a computerized glucose management system results in a substantial increase in blood glucose concentration measurements within the target range. Using a computerized glucose management system to monitor blood glucose levels, decreases in severe hypoglycemia (<40 mg/dL), clinical hypoglycemia (<70 mg/dL), and hyperglycemia (>180 mg/dL) also can be observed.
