Atherosclerotic Plaque

Atherosclerotic plaque is an abnormal accumulation of lipids, inflammatory cells, fibrous tissue, and smooth muscle cells within an arterial wall, where it can restrict blood flow and compromise vascular health. It develops when endothelial dysfunction promotes retention and modification of low-density lipoprotein particles, triggering immune-cell recruitment, foam-cell formation, and chronic inflammation; smooth muscle responses help form a fibrous cap over the lesion. Plaque biology supports the study of cardiovascular disease through tissue analysis, vascular imaging, and molecular biomarkers while guiding strategies to stabilize lesions, prevent rupture, and reduce myocardial infarction and stroke risk.

A Human Ex Vivo Atherosclerotic Plaque Model to Study Lesion Biology

Atherosclerosis is a chronic inflammatory process. This manuscript illustrates an easy to use ex vivo model to investigate fresh carotid or coronary artery plaques. The ex vivo model allows for the investigation of potential substances on the inflammatory milieu in human atherosclerotic lesions and results can be analyzed by various methods.

We describe a protocol to induce atherosclerosis in the aortic root of ApoE-/- mice fed with an atherogenic diet, through a continuous release of aldosterone. Methods to characterize plaque composition are also described.

We have developed a mechano-imaging pipeline to study the heterogeneous structural and mechanical atherosclerotic plaque properties. This pipeline enables correlation of the local predominant angle and dispersion of collagen fiber orientation, the rupture behavior, and the strain fingerprints of the fibrous plaque tissue.
