Micromanipulator Technique

The micromanipulator technique is a precision method for positioning and moving fine tools under a microscope, enabling researchers to handle cells, tissues, and other microscopic specimens with controlled accuracy. Mechanical or motorized controls convert the operator’s movements into small, stable motions of instruments such as glass needles, pipettes, or electrodes, while the microscope provides continuous visual guidance. In biology, micromanipulation supports procedures including cell transfer, microinjection, embryo and gamete manipulation, and targeted sampling. By improving control at the cellular scale, the technique helps researchers investigate development, fertilization, cellular function, and experimental responses in living or isolated biological material.

We describe how micro- and photomanipulation techniques such as FRAP and photoactivation enable the determination of motility parameters and the spatiotemporal dynamics of proteins within migrating cells. Experimental readouts include subcellular dynamics and turnover of motility regulators or of the underlying actin cytoskeleton.

Microiontophoresis entails movement of ions from a micropipette in response to a difference in electrical potential between the inside and outside of the micropipette. Biologically active molecules are thereby delivered in proportion to electrical current. We illustrate acetylcholine microiontophoresis in conjunction with micromanipulation to study endothelium-dependent vasodilation in the microcirculation.
