Rnase Omission

RNase omission is the deliberate exclusion of ribonuclease (RNase) from a biological protocol, usually to determine how RNase activity affects RNA or an RNA-dependent measurement. RNases normally catalyze the hydrolysis of RNA phosphodiester bonds, so removing the enzyme can preserve RNA or create a matched control for comparison with RNase-treated samples. In biology, this condition helps distinguish RNase-dependent RNA degradation from other sources of signal loss or molecular change. It is relevant to RNA extraction, stability studies, enzyme assays, and experimental controls, where maintaining intact RNA or verifying RNase effects is essential.

This protocol describes the 5-choice serial reaction time task, which is an operant based task used to study attention and impulse control in rodents. Test day challenges, which are modifications of the standard task, increase flexibility of the task and can be combined with other manipulations to more fully characterize behavior.

The ability to assess executive functions such as behavioral flexibility in rats is useful for investigating the neurobiology of cognition in both intact animals and disease models. Here we describe automated tasks for assessing strategy shifting and reversal learning, which are particularly sensitive to disruptions in prefrontal cortical networks.
