Mitochondrial Dna Repair

Mitochondrial DNA repair is the set of processes that preserves the genetic material within mitochondria, which is essential for maintaining cellular energy production and mitochondrial function. Because mitochondrial DNA is exposed to reactive oxygen species generated during respiration, damaged bases are commonly corrected through base excision repair: DNA glycosylases remove altered bases, an AP endonuclease cuts the resulting site, and DNA polymerase gamma and ligase restore the strand. These pathways help limit mutation accumulation and protect oxidative phosphorylation. Studying mitochondrial DNA repair clarifies mechanisms underlying aging, inherited mitochondrial disorders, cancer, and responses to metabolic or environmental stress.

Visualization of Mitochondrial DNA Replication in Individual Cells by EdU Signal Amplification

We developed a sensitive technique to label newly synthesized mitochondrial DNA (mtDNA) in individual cells in order to study mtDNA biogenesis. The technique combines the incorporation of EdU together with a tyramide signal amplification (TSA) protocol to visualize mtDNA replication within subcellular compartments of neurons.
