Ovulation

Ovulation is the biological process in which a mature ovarian follicle releases an oocyte, making it available for possible fertilization and coordinating a key stage of the reproductive cycle. It is triggered by a surge of luteinizing hormone (LH), which follows rising estrogen levels and causes follicular rupture; the released oocyte enters the fallopian tube, while the remaining follicle forms a corpus luteum that produces progesterone. Understanding ovulation supports the study of hormonal regulation, menstrual-cycle timing, fertility assessment, contraception, and reproductive disorders, while clinical detection of ovulatory patterns can guide conception strategies and inform treatment decisions.

We provide here an efficient and reliable protocol for immunostaining, Fluorescence in situ Hybridization, DNA staining followed by quantitative, high-resolution imaging in whole-mount Arabidopsis thaliana ovules. This method was successfully used to analyze chromatin modifications and nuclear architecture.
