Parasite Mutants

Parasite mutants are parasites with genetic changes that alter inherited traits, cellular functions, or interactions with their hosts, making them valuable tools in biology. These variants arise through spontaneous mutation or targeted genetic manipulation, in which researchers modify, disrupt, or replace specific genes and then assess effects on parasite growth, survival, development, or infection. Comparing mutant and unmodified parasites helps identify genes involved in virulence, host invasion, drug response, and transmission. This approach supports studies of parasite biology and host-pathogen interactions while informing the development of vaccines, therapeutic targets, and strategies for controlling infectious disease.

Genetic crosses of rodent malaria parasites are performed by feeding two genetically distinct parasites to mosquitoes. Recombinant progeny are cloned from mouse blood after allowing mosquitoes to bite infected mice. This video shows how to produce genetic crosses of Plasmodium yoelii and is applicable to other rodent malaria parasites.
