Peripheral Blood Monocytes

Peripheral blood monocytes are circulating white blood cells that support innate immunity by detecting tissue damage and potential pathogens. Produced in the bone marrow, they travel through the bloodstream and respond to inflammatory signals by adhering to blood vessel walls, migrating into tissues, and differentiating into macrophages or dendritic cells. Their phagocytic activity and secretion of cytokines help initiate and regulate immune responses. Studying peripheral blood monocytes provides insight into infection, inflammation, tissue repair, and immune disorders, while their accessibility makes them valuable for cellular assays, disease research, and evaluating responses to therapeutic interventions.

A method to expand γδ T cells from peripheral blood mononuclear cells (PBMC) is described. PBMC-derived γδ T cells are stimulated and expanded using zoledronate and interleukin-2 (IL-2). Large scale expansion of γδ T cells can be applied to autologous cellular immunotherapy of cancer.

This protocol allows for the reliable generation and characterization of blood outgrowth endothelial cells (BOECs) from a small volume of adult peripheral blood. BOECs can be used as a surrogate for endothelial cells from patients with vascular disorders and as a substrate for the generation of induced pluripotent stem cells.
