Plasmid-based Cdna Library

A plasmid-based cDNA library is a collection of recombinant plasmids carrying complementary DNA (cDNA) copies of messenger RNA transcripts from a particular cell, tissue, or developmental stage. To construct one, researchers isolate mRNA, use reverse transcriptase to synthesize cDNA, ligate the resulting fragments into plasmid vectors, and introduce the constructs into host bacteria so each colony maintains a distinct DNA insert. Because genomic introns are absent, these libraries support gene discovery, expression analysis, sequencing, and production of eukaryotic proteins in microbial systems. Library quality depends on transcript representation, insert size, and clone diversity.

We present a protocol for CRISPR-based modular assembly (CRISPRmass), a method for high-throughput construction of UAS-cDNA/ORF plasmid library in Drosophila using publicly available cDNA/ORF resources. CRISPRmass can be applied to editing various plasmid libraries.
