Plasmid Dna Transformation

Plasmid DNA transformation is the introduction of circular, extrachromosomal DNA into a living cell, most often a bacterium, to give it new genetic information. In a typical procedure, chemically competent or electrocompetent cells take up plasmid DNA through heat shock or an electrical pulse; cells are then allowed to recover before growth on selective medium, where an antibiotic-resistance marker identifies transformants. This method supports gene cloning, recombinant protein production, plasmid propagation, and analysis of gene function. In biology research, transformation provides a practical way to amplify engineered DNA and test how specific genes or regulatory sequences affect cellular behavior.

Plasmid purification is a technique used to isolate and purify plasmid DNA from genomic DNA, proteins, ribosomes, and the bacterial cell wall. A plasmid is a small, circular, double-stranded DNA that is used as a carrier of specific DNA molecules. When introduced into a host organism via transformation, a plasmid will be replicated, creating numerous copies of the DNA fragment under study.
