Protein Expression Screening

Protein expression screening is the systematic evaluation of whether, how much, and under which conditions a biological system produces a target protein. Researchers introduce candidate genes or vary factors such as host cells, culture conditions, or induction levels, then measure protein production using signals such as fluorescence, immunodetection, or enzymatic activity. In biology, this process helps identify high-performing expression constructs and optimize production before purification or downstream experiments. It supports recombinant protein manufacturing, functional studies, structural biology, and biotechnology by linking genetic design and culture conditions to protein yield, quality, and activity.

Expression of malarial proteins in cell based systems remains challenging. We demonstrate two step and one step IVT (in vitro translation) cell free expression systems for expressing malarial recombinant rhoptry proteins from HeLa cells. We use a Ni-resin affinity based purification system to purify the rhoptry proteins.
