Reversible Terminator

A reversible terminator is a chemically modified nucleotide that temporarily stops DNA synthesis after incorporation, enabling precise sequencing of genetic material. In sequencing-by-synthesis, each nucleotide carries a fluorescent label and a removable blocking group at the 3′ hydroxyl position, allowing DNA polymerase to add only one base per cycle; imaging identifies the incorporated base, then chemical cleavage removes the block for the next cycle. This controlled process produces an ordered sequence of fluorescent signals and supports high-throughput DNA sequencing, including genome analysis, mutation detection, and other applications in molecular biology.

Loading Drosophila Nerve Terminals with Calcium Indicators

Calcium is a ubiquitous messenger in the nervous system, essential for triggering neurotransmitter release and changes in synaptic strength. Here we demonstrate a technique for loading Ca2+-indicators into Drosophila nerve terminals. We also demonstrate fabrication of the required apparatus and emphasize points critical for the technique's success.

Reverse total shoulder arthroplasty is indicated for the treatment of conditions that cannot be treated with conventional arthroplasty or other procedures. These primarily include degenerative and incapacitating conditions with irreparable rotator cuff and loss of the normal biomechanical coupling of the shoulder.
