Rna Methyltransferase

RNA methyltransferases are enzymes that add methyl groups to specific nucleotides in RNA, creating chemical modifications that help regulate RNA function and gene expression. Using S-adenosylmethionine as a methyl donor, these enzymes recognize defined RNA sequences or structures and transfer the methyl group to selected bases, such as adenosine or cytosine. The resulting modifications can influence RNA stability, splicing, localization, translation, and interactions with RNA-binding proteins. In biology, studying RNA methyltransferases helps clarify post-transcriptional regulation and cellular responses, while supporting research into development, viral infection, cancer, and potential therapeutic strategies targeting abnormal RNA modification.

The present report describes an in vitro enzymatic assay to measure phosphatidylethanolamine methyltransferase activity using Leishmania cell extracts. This assay is based on the transfer of a radioactive methyl group from S-[Methyl-3H]adenosyl-L-methionine onto endogenous phosphatidylethanolamine.

This protocol describes the preparation of a synthetic tRNA substrate for the Entamoeba histolytica DNA/tRNA methyltransferase 2 (Dnmt2) homolog Ehmeth and the measure of its methyltransferase activity. This experimental approach can be used for investigating the activity of other Dnmt2 proteins.

Capture Compounds are trifunctional small molecules to reduce the complexity of the proteome by functional reversible small molecule-protein interaction followed by photo-crosslinking and purification. Here we use a Capture Compound with S-adenosyl-L-homocysteine-binding as selectivity function to isolate methyltransferases from an Escherichia coli whole cell lysate and identify them by MS.

Among different methods to evaluate gene expression, the high-throughput sequencing of RNA, or RNA-seq. is particularly attractive, as it can be performed and analyzed without relying on prior available genomic information. During RNA-seq, RNA isolated from samples of interest is used to generate a DNA library, which is then amplified and sequenced. Ultimately, RNA-seq can determine which genes are expressed, the levels of their expression, and the presence of any previously unknown transcripts.
