Section Mounting

Section mounting is the process of placing thin biological tissue sections onto microscope slides and securing them for imaging and analysis. In histology, sections produced by microtomy are transferred to a slide, positioned in a water bath or directly on the slide as appropriate, then dried and treated with a mounting medium and coverslip; the medium preserves the specimen and improves optical clarity. Proper mounting prevents folds, bubbles, detachment, and contamination, helping maintain tissue architecture during staining and microscopy. The technique supports routine histopathology, developmental biology, and research that relies on reproducible visualization of cells, tissues, and disease-associated structural changes.

Here, we describe a protocol for whole mount immunofluorescence and image-based quantitative volumetric analysis of early stage mouse embryos. We present this technique as a powerful approach to qualitatively and quantitatively assess cardiac structures during development, and propose that it may be widely adaptable to other organ systems.
