Sequencing By Ligation

Sequencing by ligation is a DNA sequencing method that determines nucleotide order by using DNA ligase to identify complementary bases, rather than extending a strand one nucleotide at a time. In a typical workflow, fluorescently labeled oligonucleotide probes hybridize to a template next to a sequencing primer; ligase joins a correctly matched probe, and imaging records its color before the probe is removed and the cycle repeated. This chemistry supports massively parallel analysis of many DNA fragments, enabling applications in genomics such as variant detection, genome profiling, and research requiring high-throughput sequence information.

MicroRNAs (miRNAs) are a widely conserved class of regulatory molecules. Here we describe a miRNA cloning method that relies upon two potent ligation steps followed by high-throughput sequencing. Our method permits accurate genome-wide quantitation of miRNAs.
