Somatic Cell Reprogramming

Somatic cell reprogramming is the process of converting a differentiated adult cell into a less specialized state, often an induced pluripotent stem cell (iPSC), by resetting its developmental identity. Typically, defined transcription factors such as OCT4, SOX2, KLF4, and c-MYC alter gene expression and remodel epigenetic marks, allowing cells to regain self-renewal and the capacity to produce derivatives of multiple tissue types. In biology, this approach supports disease modeling, drug screening, developmental studies, and potential regenerative medicine. Reprogrammed cells can also provide patient-specific systems for studying genetic disorders while reducing reliance on embryonic sources.

The protocol presented in this study describes methods for the real-time monitoring of reprogramming progression via the kinetic measurement of positive and negative pluripotent stem cell markers using flow cytometry analysis. The protocol also includes the imaging-based assessment of morphology, and marker or reporter expression during iPSC generation.

Somatic to iPS Cell Reprogramming

2023

Reprogramming alters the gene expression in somatic cells, transforming them into induced pluripotent stem (iPS) cells over several generations. Scientists can reprogram cells by introducing genes for four transcription factors—Oct4, Sox2, Klf4, and c-Myc (OSKM) by viral or non-viral methods. These factors are also known as Yamanaka factors after Shinya Yamanaka, who first generated iPS cells using mouse skin cells. Yamanaka was awarded the Nobel Prize in Physiology or Medicine in 2012 for this...

A method to generate human induced pluripotent stem cells (iPSCs) via retrovirus-mediated ectopic expression of OCT4, SOX2, KLF4 and MYC is described. A practical way to identify human iPSC colonies based on GFP expression is also discussed.

Tissue Regeneration with Somatic Stem Cells

Somatic or adult stem cells, like embryonic stem cells, are capable of self-renewal but demonstrate a restricted differentiation potential. Nonetheless, these cells are crucial to homeostatic processes and play an important role in tissue repair. By studying and manipulating this cell population, scientist may be able to develop new regenerative therapies for injuries and diseases. This video first defines somatic stem cells, and then explores the role these cells play in tissue regeneration.
