Stem Cell Injection

Stem cell injection is a procedure that delivers stem cells into the bloodstream or a targeted tissue to study or support repair, making it an important tool in regenerative biology and medicine. After administration, cells may migrate toward injured regions, engraft within suitable niches, differentiate into specialized cell types, or release signaling molecules that influence inflammation and tissue responses. Researchers investigate stem cell injection in hematopoietic transplantation, tissue repair, and experimental treatments for degenerative disease, while evaluating cell source, dose, delivery route, survival, and safety. These factors determine whether injected cells produce durable benefits or unintended effects.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

The transplantation of neural stem/progenitor cells (NPCs) holds great promises in regenerative neurology. The systemic delivery of NPCs has turned into effective, low invasive, and therapeutically very efficacious protocol to deliver stem cells in the brain and spinal cord of rodents and nonhuman primates affected by experimental chronic inflammatory damage of the central nervous system.

Intrafemoral Injection of Human Hematopoietic Stem and Progenitor Cells into Immunocompromised Mice

Intrafemoral injections allow for the engraftment of a small number of hematopoietic stem and progenitor cells (HSPCs), by placing the cells directly in the bone marrow cavity. Here we describe an experimental protocol of intrafemoral injection of human HSPCs into immunodeficient mice.

In this paper we present a method for transplanting human stem cells into various regions of the central nervous system of the chicken embryo. This provides an in vivo model for assessing the proliferation and differentiation of various types of human stem cells in embryonic tissue environments.
